The Chenango River flows through the eastern part of Greene. The Chenango River runs roughly north-south through the town. Greene is bordered by Broome County to the west and south. New York State Route 206 (NY-206), a major east-west highway, intersects with New York State Route 12 in Greene Village. The town is located on NY-206.

In 1870, the population of Greene City was 8,368. This figure includes 1,420 slaves. The city was the fourth most populous town in Missouri. Its population increased 65 percent by 1866, and it had more white children than negro children. As a result, the city and its surroundings were not threatened by 'race suicide.' In fact, the area has seen steady growth ever since.

The town was incorporated in 1882. The first settler arrived in Greene in 1792, and the first outside settlers lived in the town until 1812. It was named Hornby, but was changed to Greene after General Nathanael Greene. The town became the county seat of Greene County in 1888. There were approximately two dozen men enlisted in the Civil War, but most of them did not return.
