Located on the St. Louis, Kansas City, and Northern Railroad, Kirksville is 203 miles from St. Louis and 70 miles west of Quincy. Located on the grand divide between the Missouri River and the Mississippi, the city's water supply is best for family use, reaching depths of 15 to 20 feet.

The city is home to many colleges and universities, including the Truman State University and A. T. Still University. The town was named for Jesse Kirk, the first postmaster, who shared his turkey and whiskey with surveyors during the early days of the city. He later went on to build a hotel and a tavern.
