The city's population was 6,908 in 2010 according to the United States Census Bureau. Approximately 15.8% of residents were under the age of eighteen, 6.7% were 18-24, and 7.4% were 25-44. Another 16.6% of people were 65 and older. The median age was 44. Males outnumbered females 104.5 to 1.

The city was first settled during the United States' colonization of Florida in 1821. Its name comes from the St Marks River, which flows into the Gulf at Apalachee Bay. In 1836, the city became a hub for transportation and development as a plantation town. The railroad carried cotton to major seaports and was used for shipping. During the U.S. Civil War, Confederate soldiers were stationed in St Marks. The city suffered heavy damage during Hurricane Dennis in 2005.

St. Mark's Church was founded in 1893 by the Saint Mark's congregation. The original church was named Saint Mark's, but the congregation soon outgrew the location and built a new one. In 1893, the Saint Mark's congregation dedicated the new church building, which was designed by prominent ecclesiastical architect Carleton M. Winslow. The construction of the cathedral was delayed due to the Great Depression, but eventually it opened on December 24th, 1948.
